人臍帶間充質(zhì)干細(xì)胞 HUMSC  的詳細(xì)介紹
間充質(zhì)干細(xì)胞是特征明確的一類(lèi)**干細(xì)胞。它們有發(fā)育為成熟細(xì)胞的潛在能力而產(chǎn)生脂肪、軟骨、骨骼和肌肉。這些特性與它們發(fā)育中的可塑性共同作用,使人們對(duì)用間充質(zhì)干細(xì)胞來(lái)替換受損組織的潛在能力產(chǎn)生了極大的興趣。分離于臍帶膠樣組織(wharton’s jelly)中的間充質(zhì)干細(xì)胞,在體外含有sonic hedgehog和FGF-8的神經(jīng)元培養(yǎng)基中將轉(zhuǎn)換為神經(jīng)細(xì)胞和神經(jīng)膠質(zhì)。間充質(zhì)干細(xì)胞也可以轉(zhuǎn)化成脂肪細(xì)胞和成骨細(xì)胞。此外,在適于心肌細(xì)胞生長(zhǎng)的培養(yǎng)基中,它們有潛能分化成心肌細(xì)胞。間充質(zhì)干細(xì)胞表達(dá)基質(zhì)受體CD44和CD105,以及間充質(zhì)干細(xì)胞標(biāo)志物SH2和SH3,但不表達(dá)造血系統(tǒng)標(biāo)志物CD34。
Sciencell研究實(shí)驗(yàn)室的人臍帶間充質(zhì)干細(xì)胞是從人臍帶膠樣組織(wharton’s jelly)中分離得到的,原代凍存,冰凍運(yùn)輸。每管細(xì)胞密度超過(guò)5×10^5/ml。人臍帶間充質(zhì)干細(xì)胞經(jīng)免疫熒光鑒定、和CD73,CD90,CD105的抗體鑒定,以及分化后的脂質(zhì)染色鑒定。本細(xì)胞經(jīng)檢測(cè)不含HIV-1、HBV、HCV、支原體、細(xì)菌、酵母菌和真菌。如采用ScienCell 實(shí)驗(yàn)室特制的培養(yǎng)基,可保證此細(xì)胞15倍增增殖。
